More information please. What kind of 360 controller are you using (wired or wireless?)? Also what operating system are you using? Have you installed drivers for the controller?

To add to what mnl said, if you are using a wireless 360 controller then you must have Xbox 360 Wireless Receiver for Windows. The Play and Charge kit cable will only charge the battery, it does not take the place of the receiver.
